Is acceleration enrichment irrelevant with direct injection?
Will it also be a problem with port and direct injection?

Acceleration enrichment is largely a workaround for wall wetting issues, specifically dry walls in the intake path taking on fuel rather than it all getting into the combustion chamber, and yes DI avoids that need.
With port + DI, the concerns of PI are present. If there isn't a full wall film built up between the port injectors and the combustion chamber, acceleration enrichment or wall film compensation or similar system will have to account for that.
