I have a question on wiring harness connector. It is safe/practical to make a connector from harness branch out (i.e using DTM connector) prior connecting an oem sensors/engine socket?
If it do, how many is allowable in single cable route?

Yes you can do that. The number of connectors permitted depends on the quality of the terminals (contacts) in the connector. However, for automotive sensor use with good gold-terminal DTMs, I would not be concerned -- you could likely have 5 or 6 without issue. The higher the speed signal, the fewer connectors you should use.
Now, that does not apply to everything. For example, the USB spec doesn't permit "extension" cables, you have to basically use a device (USB Hub) to retransmit the signals.
